Inflammation is a fundamental biological process that protects tissues from infection and injury; however, chronic inflammatory signalling can also promote disease progression through sustained production of reactive small molecules such as nitric oxide (NO). Over the past decade, the Chan laboratory has focused on developing chemical biology tools to study NO signalling under physiologically and pathologically relevant conditions. My research in the Chan laboratory focuses on the development of a nitric-oxide responsive “activity-based” proximity labelling platform that can selectively capture proteins surrounding ALDH1A1, an aldehyde detoxification enzyme that is transiently inhibited during inflammatory stress. By selectively capturing proteins surrounding ALDH1A1 during inflammatory stress, this work aims to uncover the molecular pathways that link NO signaling, aldehyde detoxification, and disease progression.
